Dialysis against buffer, written by Rosa Schier

 

Buffer :  Typically buffer PBS or 150 mM NaCl, 10 mM HEPES 7.5 in 2-4L

  • 2-4L of buffer in a beaker + a magnetic bar
  • Put the dialysis membrane (from Spectra/Por) (about 15cm for 50mL of solution) in PBS 1x to get the membrane wet + clip one end (write down the name of the solution)
  • Put the solution in the dialysis membrane and close with another clip
  • Shake slowly in the fridge at 4°C overnight
  • Get the solution out and take a sample of the PBS 1x (as blank)
  • Measure the concentration A280nm on the nanodrop (2ul of solution on the nanodrop)
