Freezing mammalian or insect cells, written by Rosa Schier
- Count the cells example: 4x106cell/ml in 120ml => 480x106 cell total so we will have 48x106 cell in 10 tubes with each
- Prepare the tubes with the name and the right concentration written on it
- Centrifuge the culture 6min at 1400rpm
- During the centrifugation mix 60% of new media + 10% of DMSO + 40% of supernatant from the centrifugation. So 6ml of new media + 1ml of DMSO + 4ml of supernatant from the centrifugation in a 15ml tube and mix it well
- After centrifugation vacuum the supernatant
- Resuspend the pellet and rinse the spin tube with 9ml of the mix made before. Look at the volume that you get and adjust to 10ml if necessary
- Label all the tubes
- Put 1ml per freezing tube
- Put in the -80°C to freeze
- Stock it in a nitrogen tank
